PacBio Cuts Guidance and Pushes Cash-Flow Breakeven to 2028 as CEO Christian Henry Steps Down Mid-Transition

Pacific Biosciences delivered a second-quarter print that was overshadowed by a leadership change and a materially weaker financial outlook, even as the company touted early momentum from its new SPRQ-Nx chemistry. Christian Henry, who led PacBio for nearly six years, stepped down as President and CEO effective immediately, handing the reins to Mark Van Oene, previously the company's R&D and commercial chief. Henry will stay on as a board member and advisor. The transition lands at a delicate moment: PacBio simultaneously cut its full-year revenue guidance, lowered gross margin expectations, and pushed its cash-flow breakeven target back a full year, to 2028 from the previously communicated end of 2027.
Van Oene framed the handoff as seamless, thanking Henry for "6 years of leadership" and noting he has been working closely with the board to execute the transition. Investors will want to watch whether this is truly a smooth continuity move or a signal that the board wanted a operationally focused leader for a tougher stretch. Van Oene was explicit that his priorities are "scale is driving growth, sharpen our execution, and run a leaner, more focused organization," language that reads as an acknowledgment the company needs tighter cost discipline heading into 2027.
Guidance Cut and Breakeven Delay Are the Real Story
PacBio now expects full-year 2026 revenue of $155 million to $165 million, down from prior expectations, with non-GAAP gross margin guided to 35% to 37%, also a reduction from the range given in the first quarter. CFO Jim Gibson attributed the gross margin cut to a combination of factors: roughly $2.5 million in temporary costs tied to insourcing Vega manufacturing from a contract manufacturer, elevated compute and memory costs tied to strategic component purchases, and a slower-than-hoped SPRQ-Nx adoption curve. Non-GAAP operating expenses were trimmed to $215 million to $220 million, a $5 million reduction from the Q1 range, aided by a restructuring that will cut approximately 40 employees and save $15 million to $20 million in compensation costs next year.
Despite those cuts, the company now expects to end 2026 with $175 million to $185 million in cash, down from $236.9 million at the end of Q2 and $279.5 million at year-end 2025. Most notably, management pushed its cash-flow breakeven target out to 2028. Gibson explained the delay reflects "the continued impact of elevated compute costs and a more gradual gross margin improvement than we previously anticipated." When pressed by Jefferies' analyst on why guidance came down even as SPRQ-Nx launched on both instrument platforms and a new population-scale customer signed on, Van Oene acknowledged the tension directly: "While we do see the traction and a lot of positivity, this is in the context of the SPRQ-Nx transition... I think it's appropriate that we have this guidance range."
SPRQ-Nx Is the Product Story, But Adoption Is Taking Longer Than Hoped
The quarter's central technology development is the full commercial rollout of SPRQ-Nx, a new chemistry that allows customers to reuse each SMRT Cell up to three times, a change management says fundamentally alters the unit economics of long-read sequencing. U.S. list price per 20x HiFi human genome drops to $345, a 30% reduction versus the prior SPRQ chemistry, achieved, the company insists, without sacrificing accuracy. Over a third of the installed base had opted into the software enabling multi-use capability by June, and management expects more than half of the installed base to have adopted it by the end of the third quarter, with "the vast majority" opted in by year-end. Early yield data looks encouraging: customers are seeing "near identical" HiFi yield across the first two uses of a SMRT Cell, with only a slight decline on the third. The chemistry is also being extended to the Vega platform later in August, raising throughput to 90 gigabases per run from 60 and lowering DNA input requirements.
But the transition has a cost. Consumables pull-through guidance for the year was narrowed to $200,000-$225,000 per Revio system, as customers work through existing SPRQ inventory before ramping the new chemistry, a dynamic that hit Asia Pacific consumables particularly hard, with regional revenue down 45% year-over-year. Management was candid that the 30% price cut embedded in SPRQ-Nx needs to be offset by higher-volume usage before it becomes gross margin accretive, and that offset is now expected to show up more meaningfully in the back half of the year rather than immediately.
Clinical Validation Studies Strengthen the Long-Term Thesis
Two publications this quarter give PacBio some of its strongest third-party validation yet. A New England Journal of Medicine study, "Clinical Long-Read Genome Sequencing for Rare Disease Diagnostics," found 96.4% concordance between long-read sequencing and standard-of-care testing, with long-read sequencing improving or refining diagnoses in 3.4% of cases versus standard-of-care catching something long-read missed in just 0.2% of cases. Management argued this positions HiFi sequencing as a viable first-tier diagnostic test rather than a research tool, telling investors "a publication of this caliber in the New England Journal of Medicine also carries real weight with payers and health systems." A second paper in Nature Genetics proposed a "near-perfect genome sequencing" framework combining long-read data, diploid assembly, pangenome references and AI-driven variant interpretation, essentially building the scientific case for a one-test diagnostic paradigm that would favor PacBio's technology.
Instrument Placements Show Bright Spots Amid Funding Headwinds
Revio placements rebounded to 20 units in the quarter, up from 15 a year earlier, with 60% going to new customers and 45% part of multi-instrument orders, a detail management flagged as evidence that SPRQ-Nx economics are winning larger-scale deals. Cumulative Revio shipments now stand at 366. Vega placements fell to 26 from 38 a year ago, which the company attributed to continued U.S. academic and government funding constraints rather than demand softness, noting Vega average selling prices have normalized after promotional pricing earlier in the year and that U.S. public health labs are beginning to adopt the platform for applications like wastewater testing. EMEA was the standout region, growing 52% year-over-year and remaining, in management's words, the company's fastest-growing region, driven by hospitals transitioning from pilot programs into routine clinical production and a large multi-system Revio placement tied to a national genomics initiative. Americas revenue declined slightly, and management does not expect any near-term recovery in U.S. academic and government funding.
Reorganization Targets Marketing, Not R&D
The roughly 40-person restructuring initiated last week consolidates PacBio's marketing organization into its broader commercial team, a move Van Oene said is meant to sharpen focus on clinical customers rather than signal any pullback in core platform investment. "None of our key R&D platform projects were impacted by this reorganization," he said. Combined with the anticipated $15 million to $20 million in savings from winding down development spending on the company's high-throughput platform, management is positioning 2027 as the year operating expenses meaningfully step down, even as the cash runway assumption now extends further than previously modeled.
